i don't believe anyone's asked this

considering that twilight is the goddess of friendship and connection and all that, how does her eventually getting to "surrounded by flies" levels of perception affect her? does she just kinda mentally check out after a while?

Twilight is unique in that she stays connected to mortals! She is literally the server they use to communicate with each other / her. So she does become an abstract concept eventually, and stops participating in all the friendships that use her hair as a network, she never checks out like the other gods. The flies are precious and important, and there's MILLIONS of them so she never has to worry about taking up one pony's time. She has so many friends

Are Twilight's mane and the nature of the 'strands' allowing her to talk among thousands of mortals at once and her ability to converse with them ever at risk of degrading as she grows older, more powerful, and thus, more abstract? Or does her being innately grounded by the nature of her domain mean that she will always have an understanding of mortals, and thus, a way to reliably communicate without overloading them?

Her mane grows more powerful and more abstract. When she is connecting the entire town of ponyville, she listens in and engages with the ponies that use her mane to call each other. But when she gets bigger and starts connecting thousands of ponies, she stops paying attention and sends their communications to each other directly, instead of relaying them herself.

Twilight is unique in that she will always have that connection with mortals. She can talk with millions of them instead of just one at a time, and thus not get carried away and take up too much of one pony's time. She can also source her knowledge from her thousands of students who have personal relationships with her. This means she can get her questions answered through group brainstorming and participation. It is an honor to be chosen for twilight's caucus and contribute your knowledge and friendship for the good of ponykind.

Despite her connection, she does grow beyond mortals comprehension eventually. No pony can truly grasp her nature, but they can still talk with and befriend her.

Do gods ever miss less-serious things about being mortal? Does Twilight ever crave a good burger? Does Celestia?

Celestia has forgotten what food tastes like.

Twilight will absolutely miss good burgers when she gets too big for them. She had just perfected the perfect ordering strategy for the precise ratio of hay to tomato for maximum burger balance. She will be so sad when it's no longer feasible to order.

Though the chefs at the Hayloft will not miss her hovering over their shoulders telling them to take off two and a half strands of hay and add one beet flake or else the entire taste and mouthfeel experience will be thrown off!

But now some other pony will have to find her recipes and take up the mantle of perfect burgering. Though hopefully they take their tastes to a cookout and not a restaurant.

Since Celestia and Luna have drawbacks, would Twilight and Cadence ever develop their own?

They absolutely do, cadance especially. Cadance presides over the domain of romance, and then eventually she includes yearning, and mourning after her soul mate dies.

This is why love can be so powerful it hurts. This is why hearts break when romance ends. It didn't used to be so painful. It didn't used to take the light from your eyes. It didn't used to feel like dying. When you lost a partner you lost a partner. But nowadays you've lost part of yourself as well. The price of tying your souls together to become more than the sum of your parts means they cannot be untangled when you turn away. It means you must cut your hearts apart and do your best to stem the bleeding. You used to be able to just walk away from death, but now you must perform intricate rituals to unwind them from your soul before you lay them in the ground. Cadance has made death much more complicated, but also unspeakably beautiful.

Apologies if you've already answered something similar to this, are the gods capable of missing their old forms? Do they feel nostalgia while thinking about being able to hug somepony or eat?

Yes. Especially cadence and twilight, who were ascended by force by Celestia. They were bastions of romance and friendship respectively, before they were turned into godlings.

If they had ascended to harbinger and diety naturally, they would have had longer to accept what godhood would bring, or take steps to stop the process. Celestia and Luna do not understand where the angst comes from, because they ascended over a period of a thousand years or so. Everyone they knew as mortals is dead and gone.

They only miss the company of mortals when they slow down to look at them. Celestia felt a slight twinge of regret when twilight talked about her family and everything about them she will miss. But otherwise, Celestia has moved beyond it.

Luna misses it more than her sister.

And cadance......

Cadance can regret. She can yearn. She can wish she were mortals again and that she could nuzzle and kiss her husband like a lover should. She wants to entwine her horn with his, and touch noses. She wants to feel his breath on her lips, listen to his heartbeat. She can't, though. She can't do any of that.

She can't cradle him.

She can barely touch him

Who was the first and last of Twilight’s friends to die?

Spike was the last. Dragons can live for hundreds of years.

As for who dies first, that's an interesting question. I assume you mean of the mane 6.

Here's some possibilities.

I feel like pinkie pie would just have a random heart attack one day. Keel over. Gnight.

Fluttershy was unable to survive swamp fever, even though she helped unearth the cure, it was too late for her. The tree she became disappeared one day.

Rainbow dash breaks her neck because she tried to do a sonic rainboom while being old as balls

Apple jack dies of old age surrounded by her great grand foals

Rarity passes away of a terminal condition a few months after completing her magnum opus, which she came out of retirement to make.

So that would mean fluttershy dies first, applejack dies last, and spike outlives them all.

This is just speculation, of course. Who knows what fate really brings.

So by the time the MLP show ends, would Twilight be big enough to not interact with her friends anymore? Or does she not become a skyscraper until the last of them passes away?

And, if she does become a skyscraper while her friends are old but still alive, do they somehow visit her? Does Spike visit her?

Growing so large that ponies become bugs takes over a century. Twilight was still able to interact with her original five friends by the time the last of them passed. Here she is now!

Dragons can live for hundreds of years, so Spike was her friendship ambassador throughout the lands surrounding Equestria. After her retirement, Rarity joined him on his journeys, and the two of them visited Twilight often, before eventually flickering out themselves.

Sorry if this question has been said before, but did twilight get a cool magical girl style transformation sequence when she got her wings like she got in the show?

yes, but it was painful and confusing and she didn't know what was happening. Like getting snuck an edible and having a bad trip where you think you're dying.
