Drug discovery: A question of library design

Which approach is best for creating compound libraries for drug discovery?

This Nature review discusses fragment-based drug design and diversity-oriented synthesis.

I believe both approaches have their challenges however diversity-oriented synthesis has the scope to cover more areas of biological space.
