The Hololive DLC characters from Disgaea 6 have a Ryu number of 4, going through Etna and Morrigan to Ryu using Disgaea 6, Cross Edge, and any of the many Capcom crossovers with both Morrigan and Ryu.
My question is, would the DLC characters count as the Hololive members themselves? They are clearly intended to be; they use the same lore, and act as though they are simply inserted into the game. The characters are voiced by them, as well. However the names are slightly changed, and the appearances are altered versions of standard Disgaea 6 unit classes; Fubuki, for instance, becomes Fubuking, who is a white Prinny with Fubuki's fox ears. What is the ruling on this? For that matter, are there any other similar cases that you've encountered?
Also yes I know about Neptunia Virtual Stars as a much easier link for many VTubers, I was just thinking about the ruling on this one case.

As for the question: at least from my perspective, this strikes me as a tremendously marginal call that could be reasonably defended either way. While I have been a stickler for names, it's usually in the absence of other details that make the in-universe connection explicit.
Doing a bit of digging, if the Disgaea Wiki is to be believed, these characters are specifically the result of being Super Reincarnated, which would make them explicitly reincarnations of the Vtubers into the world of Disgaea. Complicating matters is that the in-game descriptions of the characters (which I have confirmed with footage) describe them as "forms" of the name-dropped Vtubers.
With that in mind, the only comparable and notable instance of reincarnation in video games that I can easily recall is the situation with Castlevania's Soma Cruz, and push comes to shove, I'm personally inclined to not treat Soma Cruz and Count Dracula as the same individual, as while Soma is literally the reincarnation of Dracula, they are identifiably distinct enough to be counted as their own unique individuals.
Complicating things, though, is that at least if my memory of Aria of Sorrow holds, Soma actively rejects being Dracula over being Soma Cruz, whereas I would imagine that if you were to ask, the Disgaea 6 party members would reaffirm their identity as Vtubers. But that's also of questionable relevance, since even if the bad end where he does embrace Dracula was canon, I don't think that would influence my stance on it anyway?
I'm ultimately going to come down on the side that the Disgaea 6 collab characters are the same as the Vtubers by appealing to the in-game descriptions of the characters as forms of the explicitly named Vtubers, but I certainly wouldn't begrudge someone asserting otherwise.
If you think that Manjoume actually had that much of a crush on Asuka and wasn't just making a big deal out of nothing because he couldn't deal with the realisation that he was crushing on both Asuka AND Fubuki... You can believe that, Valentineshipping is cool, but here's my take.
I like to think that Manjoume and Asuka came to an understanding after that initial duel and that they just played it up after that because they found it funny and considered it their inside joke and their brand of banter.
Lesbian Asuka helping Manjoume cope with his identity as a bisexual disaster? Her making pretend gagging sounds upon hearing Manjoume's praise of her brother? The two of them just being friends who respect eachother and don't have a weird... Stalker dynamic?
