This isn't a face of someone who wanted to celebrate the fact that they just killed The Demon. This is a face of a terrified man, who is so scared he doesn't want to think about anything. He knows he has to tell Sam, sooner or later, but he just can't face it now. So he begs, with tears in his eyes, 'can we celebrate fro a minute' wishing he wouldn't need to face the truth. Wishing he could forget that he just held his brother's dead body, that he sold his soul, that he doesn't even know if what he brought back was '100% Sam'. He is so scared he is almost crying.

Dialogue in All Hell Breaks Loose : Part Two.

For Sam, this is the most natural thing, of course he is looking out for Dean, this is what brothers do. This is what he knows about brothers, and this is a simple truth for him. it's not because Dean sacrificed for him, it's not because Dean saved his life, it's because they are brothers (remember 'What Is And What Should Never Be'). But seeing Dean's shock and confusion when he whispers 'what?', Sam realizes Dean never thought Sam is looking out for him. So he turns to a kind of logic that will speak to Dean. And so he explains why he will look out for Dean, with the reasoning he knows Dean won't have arguments for, and slowly this realization hits Dean, and in the last Gif you can see his nod, and a start of a little smile. He knows, now, that Sam got his back. And this knowledge means so much to him.

I don't think I will ever be over the complete and utter shock on Dean's face when he whispers 'What?' 

It's like this thought has never even crossed his mind. He never let himself think of someone looking out for him... Let alone his little brother.
