Kankermedicijn blijkt groei van kanker juist te versnellen. Onderzoekers doen opmerkelijke ontdekking
Onlangs bleek al uit een studie dat nieuwe ‘wondermiddelen’ tegen kanker patiënten niet helpen om langer te leven of om hun kwaliteit van leven te verbeteren.
Nieuw onderzoek heeft nu ook aangetoond dat het kankermedicijn lapatinib juist kan leiden tot de groei van tumoren.
Lapatinib wordt in combinatie met andere kankermedicijnen en chemotherapie gebruikt om patiënten met een bepaalde soort borstkanker te behandelen.
Sneller groeien
Onderzoekers van onder andere het Francis Crick Institute in Londen hebben laten zien dat lapatinib ervoor kan zorgen dat borstkankercellen sneller groeien.
“Als bepaalde borstkankermedicijnen kankercellen in het laboratorium sneller doen groeien, moeten we kijken of dat mogelijk ook gebeurt bij patiënten,” zei hoofdonderzoeker Jeroen Claus.
Ongeveer een vijfde van alle borstkankers wordt veroorzaakt doordat een eiwit genaamd HER2 kankercellen het signaal geeft om te groeien en te delen.
Andere receptor
Lapatinib is één van de vele middelen die gebruikt worden om HER2 uit te schakelen zodat de cellen stoppen met groeien of afsterven.
De onderzoekers ontdekten dat het medicijn ervoor zorgt dat HER2 zich koppelt aan een andere receptor, HER3 geheten.
Krachtiger
Samen sturen ze vervolgens krachtige signalen naar de kankercellen om zich te vermenigvuldigen, krachtiger nog dan de signalen die worden gestuurd naar cellen die niet zijn behandeld met lapatinib.

Unexpected dose-limiting toxicity in EORTC 24051: phase I study of TPF induction chemotherapy followed by chemoradiation with lapatinib in patients with locally advanced resectable larynx and hypopharynx squamous cell
In this study, the addition of lapatinib was investigated in combination with the sequential use of both approaches TPF induction chemotherapy followed by chemoradiation in locally advanced larynx or hypopharynx squamous cell carcinoma.
Seven male patients were included. Three patients were included in the first cohort, at dose level 1 (LAP 500mg daily plus TPF). Renal toxicity was observed among these three patients with 1 dose-limiteng toxicity (DLT), leading to treatment interruption. In a second cohort of four patients administering docetaxel from the second cycle, 3 more DLTs were observed. Based on the occurrence of 4 DLTs at the first dose level of LAP, patient recruitment was closed.
These results again confirm a toxicity potential of induction protocol and indicate that lapatinib cannot be combined safely with full dose TPF.
