Starting Strong: Optimizing Micropropagation Protocols for Sugarcane US-718 | InformativeBD

The sugarcane variety US-718 experimentally was carried out in changed concentration of BAP and kinetin under established condition of invitro along with replications in complete randomized design. The purpose of the study was to determine the optimum concentration in presence of combination of BAP and kinetin under invitro condition .In MS neutering media the variety US-718 shown maximum result of 90.32% shoot initiation in 3.5#0.5 of shoots per explant in concentrations of 1.5mg/l BAP without kinetin exhibited a better response than other concentration.

The present experiment was conducted to determine the ideal concentration of different plant growth regulators (BA, Kin, IBA, IAA, and NAA) for in vitro regeneration of Caladium bicolor using shoot tip explants. The work was designed in CRD with three replications. Shoot tip explants gave rise to multiple shoots when cultured on MS medium supplemented with different concentration of BA with IBA. The highest (90%) response of shoot multiplication was obtained in MS medium containing 0.25-1.0mg/L BA + 2.0-2.5mg/L IBA. The regenerated shoots were then rooted on MS medium with different concentrations NAA, IAA and IBA. The maximum frequency of rooting and highest number of roots was produced on medium containing 2.0mg/L IAA. In accordance with average growth characteristics, it was revealed that the combined effect of BA and IBA appeared to be better to individual performance. The plantlets, thus developed were hardened and successfully established in soil. The plants raised through tissue culture exhibited normal growth. Reliable protocols for micropropagation of Caladium bicolor were established, which could be used for large scale production of disease free, high-yielding, and premium quality planting material.

Micropropagation of Santalum Album L. Sandalwood

An efficient plant regeneration protocol was developed for Santalum album L. Santalaceae , an economically important species. Plant regeneration was achieved using nodal explants and leaf disc on Murashige and Skoog MS medium for direct shoot regeneration. Effect of Plant Growth Regulators PGR like 6 Benzyl Adenine BA , Kinetin KN and 2 Isopentenyl adenine 2 iP on shoot initiation 2 Isopentenyl adenine and Gibberellic acid GA3 for shoot elongation and multiple shoot formation and Indole 3 Butyric Acid IBA and a Naphthalene Acetic Acid NAA for rooting was studied. Among the explants tested for shoot induction, nodal segments proved good results. The best treatment for obtaining shoot induction was 3.0mg L BAP and for rooting 1 mg L of IBA was found to be the best treatment combination for maximum sprouting of shoot and rooting. After six the rooted plantlets were transferred for hardening, 20 of plantlets survived and resumed growth in the mixture of soil, vermiculite and sand 1 1 1 .

Abstract

In this investigation MS media supplemented with different concentrations of GA3 viz. 0.00, 0.12, 0.25, 0.50, 0.75mg/L were used for in vitro micro propagation of potato varieties Cardinal and Desiree. The parameters included were percent sprout, plantlet height, number of nodes per plantlet, number of days to root initiation, and number of days to transferable height of the plantlets. Statistical analysis showed that the varieties, GA3 concentration and varieties vs GA3 concentration were highly significant for plant height, number of nodes per plantlet and number of days to transferable height of the plantlets. Plantlets developed in M.S media supplemented with 0.25 mg/L of GA3 for Desiree and 0.12mg/L of GA3 for Cardinal produce maximum plant height, more number of nodes, reduced no. of days to root initiation and took less number of days to transferable height of the plant. Plantlets developed in these treatments were healthy and vigorous than the plantlets developed in other treatments. The results have shown that sprout percentage was high in variety Cardinal and Desiree proved efficient in response of no. of days to transferable height of the plant.
