Claire's new friends — or would-be friends — were mirrors for Dean and his perception of the way things have to be in his life. No strings. No attachment. No family.
But then Claire herself has also been a mirror for Dean, and I really really love that in the end she declared she was going to try a different way when it comes to how she lives her life. They left it sort of up in the air as to what that means exactly, but if she is Dean's mirror and she's rejecting this no attachments way of life, that is a very promising thing for what Dean's future has in store...
It’s what the humans do. They say they’re fine, and even if I don’t look it, you say I look well, and that way we avoid talking about something we can do nothing about.
10x03 // 10x09
It was made abundantly clear that Hannah was willing to go to great lengths to help Cas get better. She went to Metatron in 10x02, and nearly freed him in exchange for Cas' grace. I have to imagine she wouldn't have stopped there had Cas not been given Adina's grace by Crowley in 10x03, because while Cas thought it was something they could do nothing about, Hannah cared for him too much to sit idly by and watch him suffer.
Likewise, Cas doesn't consider Dean's condition something he can do nothing about. Dean may be resigned to his fate, he may think the only way out is to have Cas kill him if he goes too far, but there's no way Cas sees it that way. He's worried about him, he knows he has to do something, and I think right now killing Dean is going to be the very last resort for him. There is no way he will kill him before he does everything he can to save him.
How perfect would it be if this all tied together in the end by Cas' grace, the grace Hannah was so desperate for get from Metatron, turning out to be the thing that can save Dean from the mark? I keep going back and forth on whether or not I think Dean will be forced to live with the mark forever, or if angelic grace will end up being the key to erasing it. It could go either way, but regardless, Cas and his love for Dean is going to play a huge part in it all. Whether it's with his grace, or with his love, Cas is going to help pull Dean out of that darkness.

10x03 // 10x07
I've been looking at this wrong this entire time, and it just now occurred to me when I watched both of these scenes back-to-back what exactly is going on here. Sam doesn't suffer from an inability to get through the locked doors to the monsters on the other side, in fact he's the one locking them in there in the first place. But once the doors are closed, he's unable to get through to the human he knows is in there somewhere.
He thinks that by containing the monster behind a door and then reasoning with them, everything will be okay. But these monsters, they either don't want to be contained, or they're not strong enough to fight off the darkness inside. Dean chopped down the door with a hammer after Sam locked him up, while the woman that Rowena cast the attack dog spell on tried her hardest to break down the door holding her back, dying before she got the chance to break free. I think it's also worth noting that in 10x06 Olivia's dark room was unable to contain her for long, and Dean was the one to set her free.
So locking the monster up and trying to get through to them doesn't work for Sam. Is the demon cure perhaps just another locked door that will be knocked down over time? Does Sam think he's successfully locked the monster up, and now all he has to do is reason with it enough until things are back to normal again? If what happened in these two scenes is anything to go on, I would say it's looking pretty certain that the darkness inside Dean, the darkness he said he knows he can't come back from, is going to be making a return very soon. But if his brother isn't enough to contain it, what, or who, will be?

Is it just me or is Dean's left eye significantly less demonic looking than the right? There's a gray haze to it, and I can see the distinct outline of an iris. Very fitting considering all our past discussions about the left side representing the true self, and the right side representing the false self. His true self is not now, nor will it ever be, demonic. Considering this difference, and that he didn't become a demon in the traditional way, I'd say we're not going to be dealing with a typical demon. The difference in color is especially telling when you compare it to the uniform color of Sam's eyes when they turned black in Lucifer Rising.
I also can't stop thinking about Crowley telling Dean to, "see what I see, feel what I feel". How much control is he going to have over Dean exactly? I guess we have all summer to drive ourselves crazy trying to figure that out... :P
