METHODS

Bleomycin (BLM) was utilized to create pulmonary fibrosis mouse models, and TGF-β1 was employed to treat human fetal lung fibroblasts (HLFs). Then, either alone or in combination with the -catenin vector, PFD was given to treated mice and cells. In mice lung tissues, pathological alterations, inflammatory factors levels, and Collagen I levels were examined, as well as the activity of HLFs. In tissues and cells, the levels of indices associated with the extracellular matrix, epithelial-mesenchymal transition (EMT), Wnt/GSK-3β/β-catenin and TGF-β1/Smad2/3 signaling pathways were measured.

OUTCOME

The inflammatory reaction and extracellular matrix deposition in mice lung tissues were severe after treatment with BLM, which was mitigated by PFD but worsened by the addition of β-catenin. PFD decreased the activity of HLFs induced by TGF-β1, suppressed vimentin and N-cadherin levels, and boosted E-cadherin levels in HLFs, whereas β-catenin had the reverse effect. Wnt/GSK-3β/β-catenin and TGF-β1/Smad2/3  signaling pathways were stimulated in both tissues and cells, which may be inhibited by PFD.

CONCLUSIONS

PFD improved pulmonary fibrosis in vitro and in vivo by modulating the Wnt/GSK-3β/β-catenin and TGF-β1/Smad2/3  signaling pathways, which could help to improve the anti-fibrosis impact of PFD.
