Ok so now I got a pylori smoothie what do I do with it ?
Well the smoothie is a mixture cell and flagella. I don’t want the cells so I do a low speed spin in centrifuge to pellet cells and leaves flagella in solutions. Next I take the flagella solution and do a high speed spin to pellet all the flagella.
Next step (which might change in the future) i purify it though a sucrose gradient which has layers of different concentration of sucrose.
Each color is a different layer of sucrose. Sadly not usually this colorful. My student added food coloring to make the layers more visible for a picture for her presentation.
I add flagella pellet which has been dissolve in a buffer on the top and do another high speed spin overnight
The next day all the flagella will have move down to a lawyer of sucrose that has the same density as it. So that cloudy band should just be all my flagella and only flagella.

So I’m feeling motivated this week in lab again.
For those who don’t know, my main project in lab is looking to see if this efflux pump in pylori is involved in making flagella. Im order to check this it involves lots of flagella samples lol. Which leads me to my sometimes weekly 40 plates of bacteria

This is actually 39 plates lol, since one of them got contamination.

So all the cells I grew yesterday are now in the blender. I’m taking off their flagella with blender. Now they can’t swim mwahahahahah
If they can’t swim they can’t infect lol
