when Lady first met Sandor, she growled at him because Sansa was afraid:

But I keep wondering how their relationship would develop if Lady were alive? How would she communicate with the Hound? Would she start liking him at some point and how much her sympathy would confuse him?
I read someone make a really nice observation : Lady growled at Payne but despise having Sandor so near to Sansa all Lady did was "rumbling a warning". The scene IMMEDIANTLY tell us that both, LADY AND SANSA, think that Sandor is NOT a real threat.
Sansa could not take her eyes off the third man. He seemed to feel the weight of her gaze. Slowly he turned his head. Lady growled. A terror as overwhelming as anything Sansa Stark had ever felt filled her suddenly. Strong hands grasped her by the shoulders, and for a moment Sansa thought it was her father, but when she turned, it was the burned face of Sandor Clegane looking down at her, his mouth twisted in a terrible mockery of a smile. "You are shaking, girl," he said, his voice rasping. "Do I frighten you so much?" He did, and had since she had first laid eyes on the ruin that fire had made of his face, though it seemed to her now that he was not half so terrifying as the other. Still, Sansa wrenched away from him, and the Hound laughed, and Lady moved between them, rumbling a warning. (A Game of Thrones - Sansa I)
Sansa not only openly says to us that Sandor is not even half as scary and terrificant as Payne was, but that his presence and touch made her feel safe and protected in a way that only her father Ned could match, and at this point in time Sansa DOES NOT know Sandor at all.
Lady's reaction only confirmed it : while Sansa describe Payne as an overwhelming terror and Sandor as a frightning image that is way less scary and that for a moment even made her feel protected..Lady GROWLED to Payne and just RUMBLED at Sandor.
This difference is important because if Lady thought that Sandor was really a threat she would react with the same or bigger violence, yet she is calmer to Sandor than to anyone else in the scene despise Sandor being way nearer to Sansa than anyone else was and despise Sandor having his hands on Sansa's shoulders.
This scene is a freshadows to Sansa's future : all the men outside her family would be a terrible threat to her EXCEPT SANDOR.
In wolves (and dogs), growling and rumbling are related close-range vocalizations, but they differ significantly in context, pitch, and intent. A growl is a purposefully aggressive and harsh low-pitched sound and they are primarily warnings used in threatening contexts when a wolf with feels threatened, cornered and it has a protective or violent intent. In fewer words is their way to say that are ready to fight. Wolves (and dogs) also use them as a warning or threat indicating dominance or a request for space. A rumble, on the other hand, describes a low-pitched smooth sound, echoing sound, used for comunications among members of the pack. Rumbles, unlike growls, are NOT inherently aggressive and can be used for a pletora of reasons from warnings to a playfull sounds. Wolves relax their vocal tracts to produce a deep, resonant, and steady vibration to rumble while for growl they tense their vocal cords to create a rough, vibrating sound and this harshness is designed to sound intimidating and carry a clear threat.
Lady said to everyone else that she was ready to kill and die if they come near Sansa and to Sandor she said "i am watching at you man", again despise Sandor being the one touching Sansa.
Also yes : if Lady was still alive she would let Sandor pet her in the same way Sansa connect Sandor to safety.

"And what's Joff's little bird doing flying down the serpentine in the black of night?" When she did not answer, he shook her. "Where were you?"
