How is it still a debate in R&D if people are aware of the colors of mana? The first Artifacts Cycle novels (The Brothers War, etc) explicitly state they are aware there are different types of mana and that those types are tied to different types of land.

Most residents of the Multiverse are not aware of the color's of magic, but there definitely are a minority who are.

Most novels, especially from the early days, are not canon any more.

I'm having a hard time coming up with something red does strictly better than every other color. It doesn't get card advantage, it's aggressive creatures are about the same as Green after turn 1 or so, it only gets reanimation on a very small subset, etc. The one you typically cite is direct damage, but even that is often worse removal or less efficient than a creature that can block and then deal damage multiple times. What does red do that is unquestionably better than everyone else?

Direct damage is not just creature removal. It can also be used against players and planeswalkers. It is very useful and powerful and Red is #1 at it by a large margin.

Does the fact that 2/5 colors (red & black) can't deal with enchantments affect the likelihood of an enchantment matters block?

No, because we couldn't do an enchantment matters block without enchantment creatures and Black/Red can deal with those.
