Absolutely! Here's my favourite instance: the first time Taylor Scanlan got to cover Tantomile. But it's happened several times.

This week on Oasis there were several debuts and shuffles and split tracks among the male cast which meant that there were no Alonzo or Coricopat (cats who usually double as Macavity decoys—sometimes along with Gus).

So for the first time to my knowledge... Grizabella played a decoy!
