Bite the Dust
The monster under your bed isn’t a great beast, it’s a tiny dust mite. Mites’ greatest threat is for people whose overzealous immune systems develop allergies. And now a link between allergies and helminth worm infections has emerged, though whether they reduce or worsen symptoms is hotly debated. The link could come from molecular similarities between allergens such as dust mites and helminth antigens (worm proteins recognised by the immune system). To test this idea, researchers asked whether antibodies (the body’s defence molecules, red) could latch onto worms in samples from mice with no previous exposure (left), those sensitised to dust mites (middle) and those with immunity from previous helminth infections (right). They showed that dust mite sensitisation prepped the body to attack helminths, which could imply pro-allergenic properties of dust mites and helminth molecules, adding a new layer complexity to the issue, meaning the dust has not yet settled on this debate.

Past and Present
Parasitic worms, or helminths, are thought to affect over 1.5 billion people worldwide, particularly in developing nations. While rarely fatal, helminth infections can be seriously harmful, especially to children, with links to malnutrition and impaired development. In Europe, infections are now extremely rare, but evidence from helminth eggs (pictured) found in archaeological sites indicates they were common in the Middle Ages. Most recently, researchers identified eggs of both soil-dwelling nematodes and tapeworms associated with undercooked meat and fish in medieval graves in several European countries. Though rates vary between sites and species, they estimated 25% of the population on average was infected with Ascaris nematodes, mirroring some modern estimates in high-risk areas. Faced with a similar problem to affected communities today, improvements in sanitation, water treatment and hygiene processes brought infections down in Europe, before anti-helminth drugs existed, suggesting that such changes are a key part of tackling this problem.

Lethal Ladybirds
A third of the world’s population is infected with parasitic worms called helminths. Schistosomiasis is one of the many diseases they cause, resulting in 200,000 deaths a year. On a mission to find new drugs to treat helminth infections, researchers turn to insects, specifically the harlequin ladybird, which produces a range of potent anti-microbials including harmonine. They tested the effects of harmonine on Schistosoma mansoni, the helminth responsible for schistosomiasis. Growing S.mansoni adults in dishes with harmonine resulted in damage to several tissues needed for their survival, including outer protective layers, reproductive organs, and the gut. The guts of harmonine-treated helminths (pictured, right) were distended compared to untreated helminths (left), as revealed by confocal microscopy. Increasing the dose of harmonine resulted in further damage and the eventual death of S.mansoni adults. These results provide new avenues for research into novel compounds to combat the diseases caused by helminths.
