In Bloodedge Experience and XBlaze respectively we have seen the two naked and they did not have the mark at the time. Whether this is a retcon or the two gained the mark at some point in between is currently unknown.

Both Jin and Hakumen say they are unable to kill the Successor of the Azure because she has awakened as an Observer and will not be truly erased if killed by conventional means. To overcome this, Hakumen intends to make Tsubaki kill Noel using the Zero-Type Izayoi which is designed to kill Observers. However Jin absolutely refuses to accept that Tsubaki needs to sacrifice someone she must protect for the sake of the world.

The reason Izanami can’t be killed is because Noel Vermillion is binding her to this world. As long as the Successor to the Azure exists, Izanami is immortal and Doomsday cannot be prevented.
