As I've mentioned a bit before, teen Jun' is emotionally "stunted" or unstable. This comes from her upbringing where physical care was more important than mental. Jun' was favoured by the creator because she was the newest and most refined subject. Because of this he was around her more, but this doesn't mean that he was any more caring. He was often close to Jun' giving small acknowledgement in blunt, planned methods; making Jun' strive harder for his approval. The Creator used different tactics to make each of the sisters comply easier, and this affected them in different ways.
In Jun's case, she cannot control her emotions at all, leading her to look "immature" when it gets the best of her. This puts strain on her relationships and causes her to lash out when she is frustrated with herself. Her sisters never hold it against her and help her through it, but she will have to grow and learn how to cope in a healthier way herself

As for meeting the turtles: When the sisters made their escape, Jun' was left behind because of something that went wrong. (She also wasn't mentally prepared to leave anyway but that's between us 😉)
Liz, Arya, and Seraphim wandered for a while. They couldn't go back to the compound to rescue her, so they'd hoped to track her during one of her missions from the creator.
After a few weeks, the turtles caught wind of a giant and weird looking cat mutant wrecking the zoo and they check it out. Lo and behold, it's an itty kitty that turns into a giant beast the moment their back is turned. Some shenanigans later, the turtles regroup at the lair.
Thinking the turtles caught Seraphim, the sisters break in and try to get him back. They tussle a bit before they realize that he isn't there and everybody lies down.
Liz and Arya look utterly exhausted and obviously the turtles are probably curious. The sisters don't really reveal much but they do say that they are looking for their sister and Seraphim ran away while they were looking.
The Arya a convinces Liz to let them stay at the lair for a while and the turtles resolve to try and help.
While the turtles are out they see Seraphim again, only this time they follow him to the docks. The cat is sniffing around an old warehouse when the turtles jump out at him, scaring him away. Moments later, they see a figure leaping out of the building and, being so very suspicious and confrontational, the turtles immediately sense something is up and follow them.
Jun' was finishing up a collection assignment for The Creator at the docks when the turtles jump out at her. They scare her for a few seconds, but each of the sisters have a sort of biological switch in their brains that turns off things like emotions when they are in a fight. There is a moderate scale fight between them with Jun' holding her own pretty well before she throws Donnie into a rail and Raph retaliates by punching through one of her little 'water walls'. This throws her into the river and she recovers and escapes into the water. (Not without a rib injury that she is definetly getting in trouble for)

Now, the meeting takes place in between S1 and S2. Before that, the sister's lives were spent in the compound where they were created. Jun' (or "Infiltrator-15") was a result of a breakthrough, leading to expectations of higher control over surrounding areas and unique abilities. Luckily it was stopped before more were created.
Jun' spent majority of her life in a tank, across the room from her sisters (who were also in seperate enclosures). She didn't have much physical interaction with them outside of study time and group missions. This leads to an intense longing in the root of her mind to be close with them, and that never really goes away.

I've mentioned briefly before this, but Jun' has the ability to control a very specific kind of water (from the Lake of the Crying Titan) and thus carried around a gourd where a small amount was stored.
This water was used for tests and emergencies if Jun' failed to conceal herself properly. She, as her original namesake communicates, was made for stealth and infiltration. So you know the situation was dire when she needed to throw hands.
I've never explicitly stated this, only communicated through sketches and art, but in their youth, the sisters went from having collars to having Rib Circlets(basically like the colors except they rest below the chest around the abdomen) This is actually because of an incident involving Jun' getting severely injured and as a result the scientists switched to avoid damage to the subjects.
