I agree – I think it would be so much more poignant for Loki to show how much the pain really does affect him. Labor is tough and although Loki likes to brush off sentiment, I like to think he would be filled with a mix of fear and love for his and Thor’s son.
Yes, I’m thinking that portraying the labor scene as mostly serious with a sprinkling of humor will be a better choice than going the purely humorous route. It’ll keep the continuity of the story’s tone a bit more even, in my opinion. And I totally agree with your observation about Loki’s character. He’s generally very guarded, but something like childbirth would have all those emotions just bared and out there. Truly he would be at his most vulnerable, physically and emotionally. It’s a great opportunity to bolster his character and his connection with Thor and this child he’s bringing into the world.
