Alzheimer's Damage Reduced with Compound that Targets APOE Gene: Mouse Study
"Scientists have been interested in APOE for years but there are only a few examples where researchers have targeted it with a compound in living animals," said senior author David Holtzman, MD, the Andrew B. and Gretchen P. Jones Professor and head of the Department of Neurology. "Our findings indicate that APOE is not just involved in Alzheimer's risk and disease progression, but it could potentially be a real target for treatment or prevention."

Dieselben Varianten des APOE-Gens, die das wichtigste bekannte genetische Risiko des Morbus Alzheimer sind, haben in Laborstudien die Anfälligkeit von Mäusen auf eine Infektion mit SARS-CoV-2 und eine schwere Erkrankung erhöht. Teilnehmer der UK-Biobank-Studie, die beide Kopien des Risikogene auswiesen, hatten nach den in Nature (2022; DOI: 10.1038/s41586-022-05344-2) vorgestellten Ergebnissen ein deutlich erhöhtes Risiko auf einen tödlichen Verlauf von COVID-19.
Das Apolipoprotein E, das am Transport von Lipiden im Blut beteiligt ist, kann auf ungeklärte Weise im Gehirn die Entwicklung eines Morbus Alzheimer fördern, wenn sein Gen mutiert ist. Besonders betroffen sind Personen mit zwei Kopien der Variante APO E4, die bei etwa 3 % der Bevölkerung vorliegen. Bei diesen Menschen kommt es auch zu einer rascheren Atherosklerose, was einen Teil des Demenzrisikos erklärt. Außerdem ist die Abwehr gegen Krebserkrankungen und Infektionen herabgesetzt.
