the core of agnes and charlie as ani and i have figured it is that you've got a 38 year old burned out lesbian who washed out of med school and can now only work as a night shift EMT who due to cheap rent ends up living with a plucky and overachieving premed in their mid-twenties and agnes is a depressed and spineless worm with no direction who's only just barely remembering how to keep herself alive and she would sooner give herself stress-induced stomach ulcers than ever admit that she might have or even COULD have feelings for her adorable roommate that she's wound up mentoring (because that would be evil) but charlie is an extremely goal-oriented and brash individual who's gotten a crush on their usually-avoidant roommate through their mentoring relationship because she's actually just sooo gentle and soft and pretty when you get close to her like that and she also clearly gets really flustered around them when they so much as make a "joke" about this sort of thing, but logically they know that nothing could ever possibly happen between them.... unless... Ha Ha.... Unless...???
