Detecting LCA in Infants
Warren Lammert oversees Granite Point Capital, a Boston-based company that provides investment consultancy and management services, drawing on a three-decade career in the financial business. Warren B Lammert concurrently serves as chairman of Tevard Biosciences, a Boston-based company that develops gene therapies for Dravet syndrome and other rare genetic conditions.
A rare genetic disorder that is inherently autosomal recessive (meaning the defective gene must be inherited from both parents for the disease to occur), Leber's congenital amaurosis (LCA) is characterized by retinal dysfunction in newborns. In most cases, the dysfunction affects vision severely right from birth and continues thereafter, worsening at a slow pace. LCA does not cause a visible abnormality in a child's eye; the eyes look normal. However, children with LCA typically experience nystagmus (involuntary jerky eye movement) and high sensitivity to light. Infants and children with LCA also press their eyes frequently, which can cause their eyes to look sunken due to disruption of the fatty layer that surrounds the eye.
There are two gold-standard tests for LCA: electroretinogram (ERG) and blood screening. ERG exams of LCA-positive children produce flat graphs. In healthy children, on the other hand, rhythmic patterns are always present in the graph. ERG can only detect if a child has LCA but does not specify what type of mutation is responsible for the disease. According to studies, thousands of mutations can occur in the gene that causes LCA. "Mutation" is the term for an abnormal alteration in a gene.
Blood screening for LCA tests the blood for the presence of marker molecules attributed to known LCA-inducing mutations. This test often takes months to complete. On the positive side, it has an advantage over ERG since it can determine which mutation is particularly responsible for the disease if present. This vastly expands the success of treatment endeavors since more personalized approaches are enabled.
Is Dravet Syndrome an Inherited Condition? Not so much.

A past Chairman of the Epilepsy Foundation and avid supporter of epilepsy research, Warren B Lammert became interested in epilepsy activism and research after his 8-month-old daughter was diagnosed with Dravet syndrome. A rare type of epilepsy, Dravet associated with a mutation in the SCN1A gene in 80-90% of cases although the syndrome is diagnosed based on clinical as well genetic presentation. An early marker of the disease may be a long seizure episode (five minutes or longer) within the first eighteen months of a child's life which is triggered by a rise in body temperature (fever) in an otherwise healthy child. Other prominent features of Dravet Syndrome include developmental delay, speech and receptive language impairment, balance issues (ataxia) and sleep disorders. Mortality rates for people with Dravet are elevated, with some estimates in the range of 10% per decade of life. But while a genetic disorder, Dravet.is most often not an inherited condition. Some 90% of cases instead involve a de novo mutation, unique to the child and not passed on from a parent. While it can be inherited in an autosomal dominant pattern, most people living with Dravet have no family history of the disorder.
