well my headcanon is tweek bros coffee business was started by mr. tweek's dad and his dad's brother! meaning richard tweek's uncle! to explain why the coffee shop is called tweek bros coffee! because it was founded by them!
and then the brother died without having kids so tweek's grandpa passed ownership or tweek bros coffee to his son before he died later on, and mr. tweek kept the name of the business as tweek bros coffee despite being an only child!
tldr it's a family business passed between parent and child and the name refers to the original founders
