Listen, I do love Joe/Nicky a lot, but it's gotta be Wincest all the way. Sure, the sexual/romantic aspect of Wincest is purely fanon, but it IS canon that Sam and Dean are soulmates, love each other far more than normal siblings, choose each other over everyone and everything else, and have done insane things for each other, from selling his soul to almost ending the world. Their canonical brotherly relationship has everything you could want in an OTP or even real-life romance: love, devotion, sacrifice, caring, stability, domesticity, etc. On multiple occasions, they've been mistaken as a couple by strangers, discussed in terms of a couple by themselves and others who know them, and compared to real-life or in-show couples--not to imply that the brothers are actually a couple in canon, but to emphasize the intensity and/or importance of their relationship.
