I would not on the whole consider 2666 a very funny novel, in fact it might be the opposite, but it does have the funniest joke I have ever laughed at in a novel, in which a group of European literary critics, increasingly frustrated over some interpersonal entanglements, vent their anger by ganging up on and beating up a Middle Eastern (I think Pakistani) cab driver in London. Afterward, one of them shouts: "That's for Salman Rushdie!"
Catch-22 is probably the funniest novel I've ever read.
As far as movies go, the ones I remember laughing a lot at are ones I last saw when I was a teenager. I thought Tropic Thunder was really funny when I saw it in theaters. Superbad too; the quintessential "I went to a Southern California high school in the late 00s" movie.
That's as far as pure comedies are concerned. I do consider Uncut Gems to be the funniest movie Adam Sandler has ever starred in.
In terms of anime, the funniest comedy is Prison School. Elfen Lied, though, will always be the funniest anime of all time, just not for any of the right reasons. It's incredible that Elfen Lied still manages to cling to a scrap of a positive reputation (even if it's significantly diminished now compared to the 00s, when it was considered "deep" and "significant" -- seriously, go read the MAL reviews from 2009), when it is as blisteringly and obviously incompetent as Tommy Wiseau's The Room. The Gustav Klimt OP is a masterwork in that regard. When I watched Elfen Lied for the first time, there were multiple moments that literally knocked me out of my chair and caused me to roll on the floor in hysterics.
