Is it best to express to only one member, or as many as possible?
Thanks for correcting yourself. That is a trait that will take you far.
Although it depends on the dynamics of the chapter, I tend to believe it is important to at least express interest to one, and then ask them whether you should continue to express interest to everyone in the chapter. Because that first person could literally be all you need and they will tell you as much. It also might be the kind of chapter which has decided hey, we don’t need aspirants constantly telling us they are interested, just show your interest by supporting our events.
But let the first person tell you.

At my school we have spring lines, should I express before I leave for summer break or when I get back in the fall ?
EXPRESSING INTEREST DOES NOT GUARANTEE YOU WILL BE ON THE NEXT LINE OR THAT YOU MUST BE READY TO BE ON THE NEXT LINE IF ELIGIBLE.
You should express interest when you are confident that the organization is the best fit for you, that you can be a productive member and addition, and you are eligible.
So...are you going to pursue before you leave for summer or when you get back?

Is there ever a time too early to express interest?
Not really. That would be a question for yourself. If you feel you are ready, then you’re ready. No one knows how long you’ve been interested or the type of research you’ve done. I do think it is wise to wait at least a few months of the semester (if you’re a freshman) to make sure you are still interested in the org after you’ve interacted with them.

So I've done my research and I've decided to show interest in XYZ sorority. There are two things I'm concerned about.... First concern I'm not in any student orgs. The only thing I have is a 3.5,deans list,community service, and a bomb personality. I've held a leadership role as an RA but I only did that for a year. Should I be worried about not being in any student orgs? Also I don't know any members in the chapter at my school and I can be shy.. Will that hinder my chances?
1. No, participation in student orgs isn’t mandatory, it just gives members a better idea of who you are & your interests.
2. No, but it will very much help you a lot in pursuing membership. Get to know a member or two so that they know who you are aside from being an interest.

For QuietStorm: While I was expressing interest to one of the members of the org I'm interested in (AKA), she asked me what I can contribute to the chapter. I'm not sure if every aspirant for every org is asked this; but if you were asked this question, how did you know what you were able to contribute to the chapter? I know this is worded poorly lol, I apologize.
I WAS asked this question, but also prepared for it ahead of time because if I had a business (which I plan on having in the future) I would ask the same thing. Think of simple skills/hobbies you’re involved in and think of how you can apply those to an organization. When I was asked this, I mentioned that I want to be an event planner and enjoy planning events and have/had many event ideas the chapter could implement. I also brought up my background of being a military brat!
Just think of the skills you naturally have and how those can contribute to a chapter. These skills can include, but are not limited to:
Musical Skills (singing, instrumental, etc.)
Planning
Leadership
Compassion
Want to Make Change In the Community
Diverse Immersion on Campus
Social Skills
[Insert Major Here & How It Can Help A Chapter]
Whether you believe it or not, everyone has a skill set that can benefit a chapter, it just depends on how confident you are with executing and utilizing those skills, as well as if you’re willing to bring those skills to the chapters attention.
