Well, firstly, it might be just a theory, but it's possible that the creator, or rather Neko, was inspired by the play called Commedia dell'Arte, which contains the same characters. That black mark you noticed on Pierrot is a scar caused by a direct attack on Pierrot in which a lock of his hair was ripped out, There's a comic made by the creator that explains it better than I can.

Oooh That's a good question. I can tell you that we can approach this in two ways.

The first one is a bit obvious since Jester wouldn't take MC seriously because she's just a human and, like everything else, would be just another useless toy among hundreds of other toys.

And in the second, slightly more complicated, aspect would come the theme of love; without any limits, the jester would have a genuine protective interest, which contradicts all his accumulated resentment, It would be interesting to see some development on that.
