Lingual Neurofibroma as an oral manifestation of the VON RECKLINGHAUSEN DISEASE or Neurofibromatosis type 1 (NF-1). NF-1 is an autosomal dominant neuropathology that is characterized by peripheral nerve sheath tumours, neurofibromas, café-au-lait pigmentations and pigmented hamartomas of the iris (Lisch nodules). NF-1 is quite common and the familial penetrance is complete. Neurofibromin, the tumour suppressor protein product of the NF-1 gene (on chromosome 17q) is non-functional in the disease, with a consequence of uncontrolled activity of the mitogenic MAP-Kinase molecular pathway. The resulting tumours are mostly benign but malignant transformation is not uncommon. Since the MAP-K pathway is a common intracellular pathway used in cell division and proliferation, non-neural tumours (benign and malignant) may develop concomitantly.
