Just thinking the boys reactions to when Leo tells them he figured out his conspiracy theory on how/why they went through spider puberty, and who's it tiedback to. I imagine Raph has the same reaction he had back in The Evil League of Mutants episode, just taking it all in, internalizing those fuzzy memories with newfound context. Donnie, for comedic and hidden angst purposes, I would think the line from Bug Busters ("this'll teach you to compliment my work and give me my first positive reinforcement of a parent aged adult, ever!") would come back into play. Mikey, I would imagine him to try and be the most optimistic despite the weight of what was revealed intails (tho for context, I don't know if Mikey would've already started to redeem Draxum, but Imma ignore that right now for the sake of comedy and a light hearted momment in-between the angst).
[After Leo finished explaining his conclusion from his conspiracy board to his bros]
Leo: So...um... yeah we know were we got our spider quirks from...whadoya think?
Donnie: Huh, so Big Mama wasn't just a parent-aged adult, but a actually parent who complimented my work. I don't know if this makes it better or worse...
[Raph's just on his knees with a thousand mile away look on his face]
Mikey: OH! ME! GOSH!...You mean to tell us we have two dads and and a mom! What more could a turtle ask for in a family. [Mikey is just radiating with joy and sunshine]
[Leo and Donnie share a dumbfounded look]
Leo: uhh...
Donnie: ...I don't belive that's the takeaway from this, Angelo.
Leo: imma go check in with Raph.

This is really cool! Mikey is so excited to have more family and the others are like, 'did you hear anything we just said?' but the ray of sunshine they call their brother couldn't care less. All while Raph is having an exponential crisis in the corner b/c IT ALL MAKES SENSE!
but yeah, the way i have it planned to go down at the moment, is a little different than this (for drama reasons lol) but i could definitely see this happening! : )

(ps: yes, Draxum would be redeemed Lunch Lady Draxum by the time they find out. Mostly b/c Leo spent a lot of time trying to find ANY OTHER EXPLANATION that doesn't involve being the kidnapped children of one of their rogue gallery.)
