He knew all along that even if Dean was able to pull the trigger on his own brother, the Colt would have never worked. With he gun out of the way, it doesn't take Lucifer ling to over power the human. Physically Dean never stood a change and not even Sam screaming at him in the back of his mind is enough to stop Lucifer from finishing this. He'd waited long enough--hoping Michael would come out of hiding.

But he was done waiting.

Once Dean falls down, Lucifer keeps him there with his foot on his neck, pressing down hard enough to keep him from moving, but not breaking the bone. "You should have staid away. You might have survived this--" Sam's yelling was getting louder--"Sam wanted you to survive this." He says plainly and gets ready to kill the hunter.
