The a size of these sections would be determined by how secure your cylinder is against bolides and how good your external defenses are (like a form of flak or chaff or even laser deflection). Should a bolide hit and penetrate the surface of the cylinder or glass its not going to cause the cylinder to suddenly decompress like a burst balloon, you'd have a period of the before it starts to become noticeable by which time it will have been detected and a temporary repair crew could be dispatched with a permanent repair coming in behind. . Using glass as part of your shell is certainly a possibility as silicates are fairly common in the solar system. The main expense however would be making that glass secure against solar and galactic radiation. There have been many proposed solutions to this ranging from using overlapping mirrors and radiation shielding to reflect only light through such as in the second image. Concentration of light using external mirrors through a long tube in the cap of the cylinder so that it beams light in to lenses that reflect the light to the sides while any radiation that goes through is focused on the cap the opposite end like looking down a wine bottle. .
