Transgenesis is described as the stable integration, expression, and transfer of exogenous DNA into the host genome using various means like as microinjection, sperm mediation, electroporation, and so on. The melanin concentrating hormone gene was used to perform sperm-mediated gene transfer in ROHU (Labeo rohita) in this investigation (p-CMV-MCH). Milt was utilised to fertilise the eggs after being bathed in various concentrations of plasmid DNA (50-200 g / ml) for 10 minutes. The incorporation of transgene was studied via slot blot analysis. It exhibited foreign gene integration, indicating that carp sperm can pick up DNA in around 10 minutes. Furthermore, the MCH gene causes melanophores in the body, caudal region, and on the eyeball to shrink, resulting in transgenic fish that are brilliant. As a result, our research clearly shows that the MCH gene construct can be employed as a transgenesis marker gene and to investigate the molecular basis of gene integration expression and transmission.
