Can I use Duskmantle Guildmages first ability multiple times to stack it? For example I activate it twice and 4 cards enter my opponents graveyard this turn,does he lose 4 or 8 life?

Yes, the delayed trigger set up by each instance of the activation of Duskmantle Guildmage will trigger separately for each card. 

Your opponent will lose 8 life.

Let's say I have Duskmantle Guildmage and Sphinx's Tutelage in play. On my next turn when I draw, can I play Guildmage's ability in response to the Tutelage trigger? Or is that not possible because it's still the draw phase?

You can activated Guildmage before the trigger resolves, yes.

The only steps during a turn where you don’t get priority are during the untap step and the cleanup step.

Today's Arbitrary Card of the Day is Duskmantle Guildmage.

Step 1: Cast Ghoulcaller's Bell.

Step 2: Cast Mindcrank.

Step 3: Cast Duskmantle Guildmage.

Step 4: Activate Duskmantle Guildmage's first ability. Activate Ghoulcaller's Bell. Each player mills one card, triggering the Guildmage's ability. Each opponent loses 1 life, triggering Mindcrank's ability. Each opponent mills ones card, triggering the Guildmage's ability. This repeats until all your opponents are dead.

If I activated Duskmantle Guildmage's first ability, then played Mind Sculpt, would my opponent lose one life or seven life?

Seven. The Guildmage’s ability sets up a delayed triggered ability which will trigger each time a card is put into an opponent’s graveyard that turn. Mind Sculpt will cause that trigger to trigger seven times.

It’s also worth noting that each instance of the triggered ability will resolve separately, and that players can respond in between each one.
