A preliminary study published in 2015, in JAMA Internal Medicine reported that older women in Spain who ate a traditional Mediterranean diet enhanced with extra-virgin olive oil (EVOO) were less likely to be diagnosed with breast cancer by more 60%. The 3 study groups included; a Mediterranean based diet plus additional EVOO, another with the Mediterranean diet plus additional nuts and the other group being the control group. This study was originally setup to investigate the impact of diet on cardiovascular health, but also concluded significant breast cancer findings. The study ran for 5 years on 4300 women. The study needs to be re-run and designed to properly test the impact of EVOO on breast cancer. Oleocanthal, a Polyphenol in Olive Oil, Positively Impacts Human Melanoma Cells
According to new research, oleocanthal may be a potent anticancer agent for aggressive melanoma skin cancers.
One of the most aggressive, chemoresistant forms of cancer is malignant cutaneous melanoma. There are some treatment options available including injection of inflammatory molecules, along with oral and topical agents, which is where extra virgin olive oil polyphenols may play a key role.
Oleocanthal, one of the most recognized polyphenolic compounds in extra virgin olive oil, was discovered by Gary Beauchamp during an experiment to improve the taste of ibuprofen.
While researching molecular gastronomy with a team of scientists, food specialists and chefs, Beauchamp was conducting an olive oil tasting when he noticed some similarities between the taste of a particular olive oil and the ibuprofen.
Researchers have established its ability to provide potent anti-inflammatory properties, which inhibit cyclooxygenase enzymes in the same way as NSAID ibuprofen. Other studies have demonstrated extra virgin olive oil to be helpful in brain cancer chemoprevention, in reducing invasive breast cancer, in reducing breast cancer relapse, and in playing some role in the treatment of bladder cancer.
A new study, published in Nutrition and Cancer, aimed to investigate if oleocanthal could provide anti-prolific activity against melanoma cells.
A pure, direct extraction and purification of oleocanthal was obtained for the study. Human melanoma cells with high tumorigenic and metastatic activity were compared to dermal fibroblasts (normal human skin cells), each undergoing exposure to oleocanthal and tested for cell activity.
For the first time, the study revealed that oleocanthal inhibits cell growth in melanoma cells in a concentration-dependent manner, meaning the more the cells are exposed to oleocanthal, the less they proliferate and become cancer causing. The same outcome did not occur with dermal fibroblasts, suggesting the selective activity only occurs in cancerous cells.
The mechanisms for oleocanthal’s effects upon apoptosis (cell death) were studied and the results showed that the polyphenol specifically downregulates the gene expression of “antiapoptotic Bcl-2 proteins.” This in itself may provide clinical significance, as the ability to downregulate these genes can “sensitize cells to both conventional and targeted therapies.”
As for oleocanthal’s mechanisms on tumor development, researchers found that oleocanthal shows “a remarkable reduction in ERK phosphorylation and such an effect was paralleled by inhibition of AKT phosphorylation.” These proteins provide signaling pathways that form a direct communication chain that can act as an on/ off switch, which in this case appears to help switch off cancer signals.
More research will be required to support the findings but the authors suggest that given the downstream pathways, “it is conceivable that inhibition of ERK and AKT pathways may lead to suppression of cell growth.”
The authors also suggested that the activity of oleocanthal in this instance is independent of its activity against cyclooxygenase enzymes. Further studies should now be conducted to confirm if extra virgin olive oil polyphenols could, in fact, be a pharmacological agent that could be used for melanoma treatment.

Virtually all these hallmarks are targeted by dietary olive oil, particularly by virgin olive oil, since many of its beneficial effects can be accounted not only for the monounsaturated nature of its predominant fatty acid (oleic acid), but also for the bioactivity of its minor compounds, which can act on cells through both direct and indirect mechanisms due to their ability to modulate gene expression. Among the minor constituents of virgin olive oil, secoiridoids stand out for their capacity to modulate many pathways that are relevant for the aging process. Attenuation of aging-related alterations by olive oil or its minor compounds has been observed in cellular, animal and human models. How olive oil targets the hallmarks of aging could explain the improvement of health, reduced risk of aging-associated diseases, and increased longevity which have been associated with consumption of a typical Mediterranean diet containing this edible oil as the predominant fat source.

A new study has proven that an ingredient in extra virgin olive oil can kill cancer cells. Oleocanthal in EVOO causes a rupture of cancerous cells, releasing enzymes and causing cell death, without harming healthy cells.
The results of the study were made public on January 23, 2015 and published in the journal of Molecular and Cellular Oncology.
The researchers, nutritional scientist Paul Breslin (Rutgers University), biologist David Foster (Hunter College) and chemist Onica LeGendre (Hunter College) discovered in a lab study that the ingredient, called oleocanthal, causes a rupture of a part of the cancerous cell which releases enzymes and causes cell death, without harming healthy cells. In this way, cancer cells are killed by their own enzymes.
“Oleocanthal is a name for a chemical in extra virgin olive oil (EVOO) that means ‘Stinging Oil Aldehyde’,” Paul Breslin told us. “It is made by the olive when it is crushed to make the pulp from which the oil is pressed.”
“There are many compounds in EVOO that have a 6-carbon ring structure on them and collectively they are known as phenolics,” Breslin added. “These compounds are collectively good anti-oxidants preventing oxygen pore-radicals from forming and they also tend to be anti-inflammatory. Oleocanthal has been shown to interfere with processes associated with many types of inflammation, Alzheimer’s disease, and cancer formation and growth.”
The researchers discovered that oleocanthal caused cancer cells to break down and die very quickly; within 30 minutes, instead of the 16 to 24 hours it takes for programmed cell death, known as apoptosis.
Though the scientists already suspected that oleocanthal can kill cancer cells, this is the first time a study examined how this occurs.
“There are many studies that show that oleocanthal can interfere with cancer processes and growth pathways. It has also been shown in live animals that oleocanthal can shrink tumors in mice,” explained Breslin. “What is not known is whether these are all separate effects of oleocanthal on cancer or whether there is perhaps an upstream event that triggers them. We have what may be an upstream event that is a novel phenomenon to be described in that we are opening up the lysosome with oleocanthal inside the cell and releasing toxic enzymes that kill the cell. This phenomenon is called Lysosomal Membrane Permeabilization or LMP.”
“We wish to look at whether this is why tumors are shrinking in mice in the presence of oleocanthal. In our study, David Foster and Onica LeGendre focused on breast, pancreatic, and prostate tumor cells and showed they could be killed by LMP but we did not kill three kinds of healthy non-cancerous cells,” Breslin said.
Researchers would like to take the study outside the lab and investigate the effectiveness of oleocanthal to kill cancer cells and tumors in living animals.
This study confirms the anti-oxidant and anti-inflammatory qualities of extra virgin olive oil and the benefits of diets rich in olive oil like the Mediterranean diet to good health.
