The First Activity of My Apprenticeship - Explore Microbe and It's Application
Hi everyone!!! This is the first post and i'll start updating what i learned or i got from apprenticeship session. Also i don't know what do you guys prefer for language that i used, should i use Bahasa Indonesia or stay in English Language.
Those pictures were me and the crew making Paenibacillus polymyxa culture, a bacteria species act as Biological Control Agent. Paenibacillus polymyxa could secrete protease, amylase, β-glucanase, and cellulase which surprisingly inhibiting fungal diseases as antagonistic substances by disrupting the cell wall structure. Antagonistic mechanism of P. polymyxa will also form biofilm which help these bacteria becoming dominant colony. The production of siderophore and indole-3-acetic acid help plant that bioinoculated by these bacteria thrive more nicely (Yang et al., 2024).
Paenibacillus polymyxa can be cultured in-vitro using Nutrient Broth/Agar or Tryptone Soy Broth/Agar. But! If we want to use this for application in agriculture that will be expensive due to the media. We actually overcome this by making Soy Extract obtained from residential industry of soy tofu. Since filthy soy milk was a waste, we mixed with water and sugar to form medium culture with the specific ratio.
For the ingredients are:
20 liters of filthy soy milk
600 grams of sugar
4 slant isolate test tube of Paenibacillus polymyxa
All the ingredients were pasteurized at boiling point, except we don't to pasteurized Paenibacillus polymyxa, otherwise the bacteria died. LMFAO. After it has been inoculated, it must be controlled by growing in the aerobic environment, since Paenibacillus polymyxa a facultative anaerobe microorganism (Meena et al., 2017).
We make that by creating system with clean air injected using Glass wool, KMnO4 as oxidation agent or simply kill any microbe in the air. The sterilized air than enter the jerry can.
If already done fermented in 2 weeks, we can actually obtain and package in 5 liters jerry cans. These activities are new to me and makes me excited :>
