Was Rung created by quintessons? We never really got to know the true origin of Cybertronians in the IDW universe.

Like most of his fellow Selectors, Rung was a creation of humanity. The Quintesson models consisted of Slitherfangs, Pteraxadons, and the much rarer Dazzlestrike Selectors, who had no great leader like Rung to help champion their cause of freedom.

As for your second question, the genesis of the Cybertronian race is as much a mystery to us as the origin of life on your own planet. Alpha Trion would have much more to say on the matter than I, and is doubtless familiar with untold numbers of stories drawn from folklore addressing the question—stories now otherwise forgotten, displaced by the propaganda of the corrupt lineage of Primes, by the manipulations of Shockwave, by the vast diaspora caused by our own warlike actions. The very evolution of our species was shaped by the choices of so few individuals. If I had an answer to give you, you would be wise to treat it only as another yarn in the tapestry of our history.
