i find it actually so compelling in tristamp that knives sees rem as a threat to his complete ownership of vash. i know there was also tesla which was the catalyst for nai's growing resentment, and the bible, but vash's love for rem and looking up to rem is what made his hatred for humanity go to a very personal level. and i think it's interesting that because he is a plant, he has seemingly no understanding of filial relationships in the way humans do.

vash has always been more able to connect with humanity while nai struggled to understand it, in part because nai's power allowed him to be completely self-sufficient. that's why he is unable to differentiate vash's love for rem and his love for nai into things that can coexist, and love that is fundamentally different. it's just a one-dimensional scale to him from not being loved to being loved. and to him, vash loved rem more because she took care of him, and vash never relied on nai in the same way.

even so, he still offered to take rem with them because he did love vash so much that he still wanted to make him happy, knowing how much she meant to him. after she died though, and vash chose to run away from nai, his image of her warped more and more into the One Who Stole Away My Brother. then, he started thinking of humanity as the collective that stole away his brother, and he hates them all because he believes vash loves them more than him. to him it's the ultimate betrayal, and his ultimate failing.
