top/bottom discourse is so pointless. no preference is "more rooted in canon", I'm sorry. especially when we're talking about a ship that was never canon to begin with lol. "dean/sam topping sam/dean makes more sense—" well, canon dean and canon sam would never touch each other's dicks. now what?
plus, many takes on top/bottom discourse end up relying on weird psychosexual analyses and stereotypes that I don't think reflect how real people experience their sexual preferences
you can take sam and dean and form great compelling scenarios involving any kind of top/bottom and dom/sub combination. have fun !! making others feel like they "don't understand the characters" because of this preference is not cool

consensual incestuous relationships are not underexplored in fiction (even if a lot of the time its played for jokes). are you living under a rock.
When consensual incest does get depicted, it's played for laughs (as you said), shock value + as a plot twist, or framed as morally wrong and tragic.
A depiction of consensual incest as a genuine love the audience is supposed to root for and support is indeed rare, for obvious reasons. Off the top of my head there's The Borgias, Oreimo, Koi Kaze (iirc), and that's all I know about. If you're aware of some vast library of consensual incest love stories I'd love to hear about it.
But either way, abusive/noncon/traumatized incest is depicted far more often by comparison.

I love socially isolated wincest. I love that they don't know how other people work, they just know how each other work. I love them thinking the outside world is dangerous and scary and being reinforced in the idea that they only have and only want to have each other. I love dean being overly protective and keeping sam glued to his side whenever they're out in public. I love them discovering romance and sexuality together and feeling safe doing so. I love them not knowing how wrong it is to do those things with your own family. I love them being each other's brother, father, son, friend and boyfriend at the same time and not needing anything else.
