Early on I experimented with Myc's orb and how it could possibly animate when talking. We ultimately went with its original simple ball, but it was fun to mess around with. Jeffrey Thompson directed me towards bio luminescent wildlife. I also thought of throat and brain anatomy, and a floating ball in liquid.
