Cassandra had been following Desmond for at least 10 minutes, carrying a rather large box, waiting for him to notice her following him.
Lack of training or not, Desmond had picked up on when people were being less than coincidentally in his same path. So once he hits a relatively bare area, he stops and gives a long sigh, hands in his pockets as if this is normal.
