The thing about Desmond is that he never just got to live his own life, and I mean that literally. He spent his entire youth training against am enemy he never saw, and when he eventually ran away he had to spend the whole time moving from place to place, always looking over his shoulder, never free to just be.

Then, of course, he was captured by Abstergo, where he sees some of the life of Altaïr. Then, he joins the assassins, where he experiences practically the entire life of Ezio. And then the same again for Connor! He spends dangerous amounts of time in the animus, and just enough time in the real world to kill someone he trusts, get messed up by the bleeding effect (because the other lives he experienced now follow him into his own), and learn that he has to die.

Desmond's story is tragic because he never had his own life or his own freedom, and even after his death only a handful of people will ever know of the sacrifice he gave.
