If Two-Face’s personalities had somehow been separated into two different bodies, how would Riddler react? Would he try to put them back together?
That would depend on the nature of the separation. Harvey and Harv are halves of a whole, each accounting for different facets of one personality. Both sides of Two-Face have tried to get rid of the other, only to discover that they are, in fact, one guy. They fit together and have always needed each other. With this in mind, there are two major ways they could be divided, which we can call an organic split and an absolute split.
The organic split assumes that each of the two different bodies has one permanently dominant personality and one subdued one that each add up to a complete person, able to function and take care of themselves. Something akin to adapting to the loss of one of the senses. In this scenario, Harvey would likely attempt to return to his old career and life before his accident, and Harv would run amok. They would part ways and keep their distance from each other, which would be a net loss for Ed. Both Harveys would still like him, but their hatred for one another would result in an awkward and unpleasant situation all around. Something something divorce leads children to places.
The alternative is an absolute split, in which Harv and Harvey just divide down the middle into two bodies. In this situation they would have to remain codependent, as neither has the adaptive tools to exist alone. They would lose access to each other's thoughts and the built in empathy of sharing a body, which would make their already uneasy relationship more of a struggle. While Ed might enjoy the idea of having two Harvey Dents to fawn over him, he wouldn't actually like watching them suffer and would look for a way to recombine them.

The Judge is a strange entity. He is a manifestation of the guilt Harvey feels for failing to live up to his own morals and a further splintering of his psyche, separate from his usual duality. He does not appear often and took Ed by surprise after he'd already known Harvey for some time.
Initially, Ed was terrified. The Judge could not be reasoned with or soothed using any of the tricks that work on Two-Face. He ignores the coin and Harvey's other compulsions and, because he does not identify himself as Harvey Dent, he did not recognize any familiarity with Ed. To the Judge, Edward was the Riddler, a guilty party that needed to be punished.
After a few life-threatening encounters, Ed came to understand that the Judge adhered only to legal procedure and valued impartiality above all else. Ed had to prove, in court, that his marriage to Harvey also applied to the Judge, making the latter's retribution against him a conflict of interest. Once the case was made, the Judge instantly calmed down and became a docile, surprisingly gentle being.
No longer endangered, Ed now sees the Judge as yet another intriguing facet of his ever-compelling Harvey.

Harvey is definitely getting Edward a second slushie. He never allows anyone in his circle to have only one item on their person, it must always be two. Plus, it gives him an excuse for him to get in line a second time. A major double win for Harvey!

I haven’t really had the opportunity to play games for a while, but the customs in Tomodachi Life look like so much fun! If I do get it someday I would definitely make Ed and Harvey and company.

Do Two-Face and Riddler get irritated with each other’s compulsions? For example, Two-Face backing out of plans at the last minute or causing destruction and plans to go awry because the coin told him to, or Edward insisting on leaving riddles at crime scenes even if it risks getting them thrown into Arkham or getting Ed hurt
Absolutely. They also relate to each other's impulses differently. For Ed, the RNG is what he signed up for. The coin flip is a part of the game. He gets frustrated when Harvey makes him lose but accepts it as an outcome. He's less taken with the destruction if he's anywhere in the line of fire, but finds it delightful otherwise.
Harvey doesn't like riddles, but he likes to see Ed enjoy himself. When it comes to the liability of crime scene riddles, he's split between being furious and being excited, because it greatly raises the chances Batman will show up.
Ed getting hurt is a risk no matter what the nature of the heist. Harvey, a bruiser through and through, initially wasn't used to having a smaller and more delicate partner in crime. Despite Ed's single hit point, he is surprisingly wily and good on the offensive. He also values his skin and tends to stay out of the way of physical danger. Over time, Harvey came to accept the way Ed operates.
Harvey has mixed feelings about going back to Arkham, and about Ed being there. Part of him still strongly believes in punitive correction and figures it a natural consequence to their impulses, and part of him despises all of it.
