After a more thorough reading while re-reading in Japanese I have come to the conclusion that Vash's mysterious ailment in volume 2 chapter 6 of Trimax (Home Sweet Home) is just exhaustion. It's said they've been taking turns driving without stopping, because they're trying to get to the colony ship after Rai-Dei informed them of it being targeted, but the implication is that Vash ended up driving for too long on his turn(s) and drove himself to exhaustion. Silly boy.
