It depends on what type of poison it is.

Man-made poisons like some pesticides will breakdown and become less poisonous.

Other poisons like certain herbicides break down into dioxanes and become more poisonous.

But, I'm assuming you want to know about the fancy stuff like arsenic. The problem here is that expiration makes it less effective but more poisonous. So the poison becomes harder to use or may take longer to kill but, the dosage necessary to do it is lower.
